Domperidone Tablets

Descriptions : Domperidone is a peripheral dopamine antagonist (blocking D1 & D2 receptors).

Actions : Domperidone facilitates gastrointestinal smooth muscle activity by inhibiting dopamine at the D1 receptors and inhibiting the release of neural acetylcholine by blocking D2 receptors. It does not cross the blood-brain barrier nor the placental barrier but exerts its effects at peripheral sites: the chemoreceptor trigger zone (in the brainstem) and the gastrointestinal tract. It is less likely to produce extrapyramidal reactions or drowsiness.

Pharmacokinetics : Bioavailability 13-17% ( due to the "first-pass" hepatic metabolism and gut wall metabolism).Time to peak concentration is 30-110 minutes .Total protein binding is 91-93%. Elimination half-life is 7.5 hours. Excretion from renal is 1.7-2.4% and feces 10%.

Indication : nausea, vomiting, postprandial dyspepsia.

Dosage and Administration : Oral, 10-20mg tid-qid taken 15-30 min before meals.

Adverse drug reaction : dry mouth, transient skin rash, headache, thirst, abdominal cramps, diarrhea, drowsiness, nervousness.
